How to reach Sissu Valley from Manali: Reach Manali from Delhi or Chandigarh. You can either take a bus or taxi to Keylong. Cross the Atal Tunnel and Sissu is the first stop after that.

How to reach Sethan Valley from Manali: Take a taxi or a bus; the ride from Manali to Sethan takes roughly an hour; taxi fares are ~1000 INR and bus fares are roughly between 150 to 1100 INR.
